The morpho-functional state of Vicious-Circle target organs in mice with the induced of citokindependent hepatitis |

The
model induced by Con-A is received for the investigation of basic
processes of liver pathology, initiated by activation of lymphocytes,
infiltration and characterized by DNA fragmentation, significant release
of transamins and apoptosis. A number of studies showed that the release
of cytokines by activated cells leads to the increase of NO through
inactivation of NO-syntetasys, this can result in a negative hipotropism
and myocardial injury. Therefore the simplicity of the mentioned
experimental model fully satisfies our theoretical and practical needs on
this stage of investigation. The
experiment was hold in 30 mice of pure line, weighing 150-200g, which had
been in quarantine. The first group (20 mice) - 2 ml of Con-A were
injected. The second – control group (10 mice) interperitoneally
received physiologic solution (the doses calculated per 1kg. of weight).
The experiment was hold in surgical department of the laboratory,
observing all rules of aseptic and antiseptics were strictly observed.
After 24 hours post Con-A injection the animals were decapitated and
samples were taken from their heart, brain, liver and blood. Free-radical
processes and NO were studied using spectrometric resoner with quartz
diurer. The computer program and self -registration device were receiving
signals of paramagnetic spectrum that were registered by spectrometer PE
1307.
The morphological changes on liver, brain and heart were studied by
electronic and light (luminous) microscope. For measurement of contractile
ability of contractile protein system we used glycerized heap (sample) of
fibers obtained from myocardium of right and left ventricles The
study of paramagnetic spectrum revealed the strongly developed dysfunction
in all the studied organs, the release of Fe contributes to inactivation
of prolipherative processes, the high concentration of Mn+2 ions
indicates to the inhibitive activity of adenociclasies which in its turn
initiates inactivation of peroxides and also is the reason of
adrenoreceptorial inactivation of the
blood elements. The appearance of paramagnetic signals of Mo+5
containing xanoxsidaze indicates the existence of the obstacle for
transportation of elements in mitochondrial energetic function The
function of a whole body as well as the function of the separate organs in
the certain period – (during borderline state), are stable. Whenever in
target organs conditions supporting pathological state develop. The
general reactivity of body decreases, occurs damage in the balance between
oxidative and antioxidative systems and the influence of the initiating
factors contribute to development of vicious-circle which determines the
gravity of pathological disorders in target organs, and their variety
(destruction in brain, heart and liver are strongly developed in parencim
as well as in microcirculation system. There are sighs of vacuole
dystrophy, granular dystrophy and leikocytarial infiltration of the liver,
sighs of atrophic changes of cardiomiocits in heart that are characterized
by destruction of miophibriles and mitochondria and finally in the
gliocits of brain anoxia- ischemic occur).
On
the basis of the mentioned changes in comparison with the control group,
the well expressed decrease
of contractile function of the left and right ventricle (38.8% and 46.8%
respectively, p< 0.005) can be considered as a serious disorder. The
results of an experimental study confirmed the existence of
pathomorphological changes registered in our clinical and authopcial
studies of the materials obtained from patients with hepato viruses. We
assume that considering predictable morpho-functional complications of the
main disease as well as the separate organs - borderline state -
especially the viscous circle target organs in development of viscous
circle will enable us to start preventive treatment on the earlier stage
of dieses.
